Luminex Multiplex Assays
Luminex 100
The Luminex 100 platform is used to acquire data from multiplexed assays using a capture bead system developed by the Luminex Corporation. An excellent tutorial explaning how the Luminex system works can be found at the corporate website www.luminexcorp.com Based on a dye color coded scheme up to 100 separate tests can be conducted on a single drop of plasma, serum, lysates or other biological samples. Each bead set will have a separate capture reagent attach to the surface that is directed against a single analyte. Depending upon what analyte is being screened one to several bead sets are incubated with the sample in order capture the analytes. Because of the special dye ratio incorporated each bead, each unique bead population can be analyzed separately after acquisition. Currently, we are screening for cytokines and chemokines from human and mouse samples although the procedure is not limited to theses analytes. The bead populations are detected using a 635 nm laser and they are identified based on the ratio of proprietary dyes found in the beads. The captured analytes are detected using a 532 nm laser which is used to excite the fluorochrome phycoerythrin (PE) that is attached to a detection reagent. In the case of cytokines and chemokines a PE labeled antibody directed against the analyte is used as the detection reagent.
Multiplexing allows for multiple tests per sample thereby reducing the amount of sample required for screening. A standard curve is generated for each cytokine and chemokine used in every assay to ensure accurate data. For pricing information, please contact the lab at (716) 845-8471 or email us.
