MacIntosh – CellQuest Pro  “General Instructions” for the FACscan

1. Log in to your account.  An account will be setup on the hard drive for each user.

2. On the desktop double click on the “CellQuest Pro” icon. A blank window should appear.  Select “Acquire” from the Title Bar and then select “Connect to Cytometer” from the last item on the menu. The Browser window should automatically pop up. 

3. If you do not already have a template created, use the Tool Bar box (left side of the screen) to select the dot plot icon.  Using the mouse draw a new dot plot window in the blank window.  The Inspector window will then pop up on the screen.  In the Inspector window goto the top selection and change this “acquisition”.  Select the X & Y parameters for the plot.  Click OK.

4. To add more plots or histograms  of the same size select “Plots” from the main Title Bar.  Make sure Plot source is set for acquisition.  Select the X & Y parameters for the plots. Acquistion windows (templates) created by the user can be saved for future use.  Under “File” located in the main Title bar select “save document as” and save the windows settings to your folder.  These templates can be opened under “File” from the Title Bar after opening CellQuest.  Just select “Open”.

5.  To obtain the windows for Detector/Amps, Threshold and Compensation select “Cytometer” from the Title Bar. Alternatively, hold down the “apple” key and then press the “1”, “2” and “3” number keys.

6.  To obtain the Counter window select “Acquire” from the main Title Bar.

7.  In order to set the acquisition conditions select “Acquire” from the main Title Bar and then select “Acquisition and Storage”.  Input selections.

8.  To set the File Storage conditions goto the Browser window.  If the Browser window is not present then select “Acquire” from the main Title Bar and the select “Parameter Description”.  Set the destination folder (your folder) and file name.  Select the top “change” button across from directory to create a folder. Use only alpha numeric characters.  In the pop up window select your folder destination and name.  Click “create” and check the file path.  If the file path is okay then select “choose” to use the new folder. To create a file name select the bottom “change” button.  A new pop up window appears. In the top line create the filename (it is recommended that you use your initials and the date).  At the bottom of the pop up window reset the file count.

 

  • Instrument Settings:  Initially, you will manually set on the cytometer or on the computer screen the settings for each experiments.  All settings used will be automatically saved to each file collected and these can be recalled for future use.  It is recommended that each user save their settings to a file on the hard drive as each instrument uses unique settings. To save your settings go to the main Title Bar and select “Cytometer”.   Next select “Instrument Settings” – “Save”.  Select the destination folder and type in a name for the settings file which includes the word “setting” Click “save” then “done” to finish.  To recall settings saved on the MacIntosh, select “Cytometer” from the main Title Bar.  Under this heading select “Instrument Settings”.  In the instrument settings window select “open” and find your files.  Double click on the file or select the file and click “open”.  After selecting the file, click on “set” to load the settings to the cytometer and then click “done”.
  • Go to the Main Title bar and select “acquire”.  Select “counters” to open the counter window.

 

  • In the Browser window leave the “setup” box checked until the instrument is ready for sample acquisition.
